ThetreatmentofMYCN2(±Dox)andBE(2)-CcellswithGC7Sulfatefor72hatvariousconcentrations(0.1to100μM)significantlyreducesthenumberofviablecellsinadose-dependentmanner.InMYCN2cells,5μMofGC7Sulfateinhibitscellviabilityby~40and~60 percent ,respectively,comparetountreatedcontrolcells.BE(2)-Ccellsrequire25μMofGC7Sulfatetoreducecellviabilityby~ 50 percent .Exposureto10and100μMGC7for72hclearlydecreasesthelevelsoftotalretinoblastoma(Rb)andphosphorylatedRbaswellasofCdk4prChemicalbookotein,andincreasesthelevelsofp21protein.Between0and20μM,GC7SulfateinduceslittlecytotoxicityinHCCcells,whilehigherconcentrationsofGC7Sulfate(50to100μM)significantlyinhibittheviabilityofallfiveHCCcelllinestested.Newlysynthesized3H-labeledhypusineofeIF5A1/eIF5A2israrelydetectedafter20μMGC7Sulfatetreatment,comparetountreatedcontrol.Theactivityof[3H]-spermidineincorporatedintoHCCcellsissignificantlydecreasedby20μMGC7Sulfateorhigherconcentration.
